Abstract

Polymorphisms in the ACTN3 gene affect the ultrastructural morphology of sarcomeres that can influence fiber type distribution. The predominance of specific fiber types in lower limb muscles might improve performance in endurance or explosive events. Recent data shows that homozygosis for the R577X allele of the ACTN3 gene leads to greater performance in endurance events while R577R homozigotic individuals perform better in explosive events. Concurrently, it is known that responsiveness to postactivation potentiation (PAP) in influenced by fiber type distribution with type IIx fibers responding the most to pre-activation. The aim of this study is to investigate the influence of ACTN3 polymorphisms on responsiveness to PAP. Our hypothesis is that R577RR individuals respond better to PAP due to greater proportions of type IIx fibers. Untrained males will be assigned to two groups based on their ACTN3 polymorphism homozygosis: XX and RR. Their countermovement jump height will be determined in two distinct conditions: control (no pre-activation) and potentiated (four minutes after performing five squats with a 5RM load). Different conditions will be randomly performed in separate days. All participants will undergo as many familiarization sessions as necessary to achieve reproducible countermovement jump height before fata collection. Differences between conditions and groups will be compared by mixed-design factorial ANOVA (groupsvs conditions). Tukey´s post hoc tests will be performed upon identification of a significant interaction effect. Significance level will be set as p < 0.05.
