Abstract

The genome of Salmonella Typhimurium encodes a type VI secretion system (T6SS) inside the Salmonella pathogenicity island (SPI-6). In Salmonella Gallinarum the T6SS is encoded from SPI-19. Genes located inside SPI-6 T6SS from S. Typhimurium are activated during late stages of infection, while SPI-19 T6SS from S. Gallinarum are activated early during the infection of macrophages. The significance of the T6SS activation under these conditions is still unknown. To obtain further knowledge about the activation dynamics and the assembly of SPI-6 and SPI-19 T6SS, we plan to make bacterial strains expressing a structural T6SS component in phusion with superfolder GFP (sfGFP) to follow at the single cell level its assembly during the infection of different eukaryotic cell lines. At the end of the project, we expect to have a broader and integrated view about the activation of T6SSs during the intracellular stage of infection.
