Probiotic for black pacu (Colossoma macropomum): effects on productive performance, survival, hematoimmunology and intestinal evaluation

Abstract

The black pacu Colossoma macropomum is the second most produced fish species in Brazil, but it does not have a well-developed technological package and end up suffering with problems inherent to aquaculture. The use of probiotics has been encouraged by its effective prophylactic method that improves intestinal health, immune response, nutrient uptake and productive performance of the host and even the quality of the water. However, until date, there is no autochthonous probiotic for tambaqui. To develop a probiotic for this species, autochthonous bacteria (Enterococcus hirae, Lactococcus lactis, Pediococcus pentosaceus and Staphylococcus hominis) were previously selected by our research group, due to the promising in vitro effects. These strains will be lyophilized and added to the feed at 108 CFU/mL and offered for 60 days to the fish. The productive performance and survival to Aeromonas hydrophila infection will be evaluated and the bacterium which presents the best result in this experiment will be selected for a second study which will be added to the feed at concentrations of 107, 108 and 109 CFU/mL for 60 days. On days 0, 7, 15, 30, 45 and 60 the hematological, immunological and biochemical parameters, intestinal morphology, and quantification of probiotic bacteria in the intestine will be analyzed by real-time PCR. With this project, we intend to select a probiotic bacterial strain and determine the dose and period for its administration, characterizing its immunostimulatory effects on the health of the host.
