Effect of red wine consumption on the induced Apical Periodontitis development in rats

Abstract

Wine consumption is a common habit worldwide, being one of the main sources of polyphenols through the diet; among them resveratrol and quercetin, demonstrate osteoimmune-protective activity in the host. In addition, the low alcoholic concentration present in red wine, combined with light consumption, also contributes to the regulation of bone balance. However, there are no studies that relate the compounds present red wine with endodontics and more specifically, the systemic action of these on Apical Periodontitis. The objective of this study will be to analyze and compare the development of Apical Periodontitis in rats with normal diet or diet supplemented with red wine consumption. Thiry-two Wistar albino rats, all receiving the Apical Periodontitis induction, will be allocated in 3 groups as follows: Control (C) normal diet; Red Wine (VT) administration of 4.28 mL/kg of red wine via gavage once a day; Alcohol (ALC) administration of an alcohol solution at 12.5% via gavage once a day; and Resveratrol + Quercetin (RESV + Q) administration of a solution containing the same amounts of these compounds present in red wine via gavage, prepared with Resveratrol (R5010 - purity e99% - Sigma) and Quercetin (Q4951 - e95% purity - Sigma). Throughout the experimental period, the animals and the feed consumed will be weighed daily. After 30 days of injury induction and administration of the diets, blood collection contents will be performed, after which the euthanasia of the animals will be performed in order to analyze and compare micrographically and histometrically the periapical lesion; analyze and compare the tissues of the periapical region of rat teeth using the expression of the TRAP, OPG and RANKL proteins as cellular indicators of bone resorption; analyze and compare the tissues of the periapical region of rat teeth using the expression of cytokines and IL-1², IL-10 and TNF-± as indicators of local inflammation; analyze and compare the expression of IL-1² and IL-10 cytokines as indicators of systemic inflammation; analyze and compare the neutrophil, eosinophil, lymphocyte and monocyte concentrations among the experimental groups by hematological analysis; and to analyze and compare the concentrations of the cellular activity markers of bone metabolism: calcium, phosphorus and alkaline phosphatase.
