Proteomic analysis of acquired enamel pellicle in patients with head and neck cancer submitted to radiotherapy

Abstract

Cancer is one of the leading causes of mortality worldwide and is currently a major public health problem. The acquired pellicle (AP) is a tegument formed in vivo as a result of the selective adsorption of salivary proteins on the surface of the tooth, which also contains glycoproteins and lipids. The presence of proteins in AP forms a protective interface on the surface of the tooth, participating in all the interfacial events that occur in the oral cavity. It is an organic film, free of bacteria, that covers the dental tissues. Works have focused on the characterization and protective impact of the acquired pellicle formed on the enamel surface. However, little is known about the changes in the composition of the acquired pellicle in patients with head and neck cancer submitted to treatment by radiotherapy, which interferes with the amount and quality of saliva. Based on this, the objective of this study will be to evaluate the protein profile of the acquired pellicle in patients diagnosed with head and neck cancer and submitted to radiotherapy treatment. Participants will be 18 people, aged over 18 years, 9 participants without diagnosis of head and neck cancer, non-smokers, good general and oral health and 9 others diagnosed with head and neck cancer submitted to radiotherapy. The acquired pellicle will be formed in the morning for 120 minutes after prophylaxis with prophylactic paste. Acquired pellicle formed will be collected using filter paper soaked in 3% citric acid. Samples will be processed for nLC-ESI-MS / MS analysis. The obtained MS / MS spectra will be compared with human protein databases (SWISS-PROT). The free quantification of markers will be performed using the PLGS software.
