Proteomic analysis of periodontal ligament of rats submitted to experimental periodontal disease induction and obesity

Abstract

Studies have been conducted to better understand the effects of periodontal disease on periodontal tissues. However, there is no consensus in the literature about these effects on the periodontal tissues of obese individuals. The objective of the present study is to evaluate in vivo the effect of obesity on the progression of periodontal disease. A total of 40 rats were distributed in 4 experimental groups: control group, without intervention (C); induction of obesity (O); induction of experimental periodontal disease (P) and induction of obesity followed by induction of experimental periodontal disease (OP). The animals were sacrificed after 21 days of onset of periodontal disease induction. The protein composition of the periodontal ligament of rats submitted to the induction of experimental periodontal disease, associated or not to obesity, will be evaluated through proteomic analysis. From this analysis, 5 proteins with significant differences between the groups will be selected, with relevant function for the inflammatory process, to validate their presence in the periodontal ligament by means of immunohistochemistry. The gene expression of these proteins will be evaluated in rat gingival tissue by means of RT-qPCR. All data obtained will be evaluated, and then submitted to a normality test. From this, appropriate statistical tests will be used for comparisons between groups. In all procedures, a significance level of 5% will be adopted for the decision on the validity of the hypothesis tested. (AU)
