Evaluation of topical Ozonotherapy in cutaneus grafts in the treatment of surgically induced wounds in Wistar rats

Abstract

The skin graft consists of transferring a segment of cutaneous tissue from the donor area to the recipient bed. Ozone therapy is defined as being an oxidative therapy that generates an oxidative stress, and consequently an antioxidant effect on the body. There are no citations in the literature regarding to experimental studies of the action of ozonated sunflower oil and ozone gas on skin grafts. Thus, the present study aims to evaluate the topical ozoniotherapy with ozonated sunflower oil and ozone gas in autogenous skin grafts in the treatment of surgically induced wounds in Wistar rats by means of macroscopic and tomographic evaluation of the grafts; histological evaluation of grafts and evaluation of tissue oxidative stress. Thirty-six healthy Wistar rats, males, 18 weeks years old, and body mass between 300 and 350 grams will be used. The rats will be divided randomly into four groups of nine animals: Group 1 - rats submitted to wound induction and autogenous graft and treated with saline solution (0.9%) topically every 24 hours; Group 2 - rats submitted to the induction of wounds and autogenous graft and treated with ozonated sunflower oil (2 mL) topically every 24 hours; Group 3 - rats submitted to autogenous wounds and graft induction and treated with ozone gas subcutaneously every seven days; Group 4 - rats submitted to the induction of wounds and autogenous graft and treated with ozone gas subcutaneously every 7 days, and ozonated sunflower oil (2 mL) topically every 24 hours. After the anesthetic procedure two wounds of 4 cm in diameter will be performed and autogenous grafting will be performed on the dorsal thoracic region. The variables to be evaluated will include: macroscopy and microscopy, tomography and oxidative stress. Analysis of variance (ANOVA) for repeated samples, with contrasts by Tukey method and Kolmogorov-Smirnov test will used. Data that do not meet the normality characteristics will be used Friedman test. For comparison of the four groups will be used analysis of variance for independent samples (ANOVA One way) for normal data, and Kruskal-Wallis test, for data outside the conditions of normality. Differences will be considered significant with p  0.05.
