Evaluation of resin infiltrant modified by bioactive nanofibers in the treatment of caries-like lesions in bovine enamel

Abstract

With the understanding of dental caries as a dynamic process, early detection of the lesions allowed less invasive treatments. Among the strategies for the treatment of active white spot lesions, the resin infiltrant has been highlighted, however, a major limitation of the technique is the inability of the adjacent enamel remineralization, as it only paralyzes caries lesions. Thus, the present research aims to evaluate the anticaries potential of a bioactive resin infiltrant previously developed by this research group. Firstly, 3D hybrid cotton wool-like nanofibers (100 nm) of Poly-lactic acid (PLA) doped or not with bioactive ions were incorporated into a resin infiltrant (ICON, DMG). In this project, bovine tooth enamel blocks (6 × 4 × 2 mm) will be prepared and selected by initial surface hardness. Then, the blocks will be submitted to a demineralizing solution for induction of caries-like lesion and treated with one of the following groups: 1-ICON (control), 2-ICON + PLA/ Ca3(PO4)2 and 3-ICON + PLA/SiO2 (n=12/group). After protection of 1/3 of the exposed surface, a new third will be submitted to a pH-cycling regimen (DES/RE), for 7 days at 37 ° C, in order to evaluate the resistance of the infiltrant in a new cariogenic challenge. The analysis of the variables will be conducted in all conditions (sound, after caries induction, after treatments and after DES/RE challenge), considering surface hardness and X-ray dispersion energy spectroscopy (EDX). The quantitative data will be submitted to two-way ANOVA, followed by Tukey test, with a significance level of 5%.
