Effect of fluoreted solutions associated with copylimer aminomethacrylate on the antierosive potential of the acquired film: in-situ randomized crossover study

Abstract

The frequency of individual presenting tooth erosion, especially children and adolescents, has increased significantly over the years. The evolution of the condition over the years can result in tissue loss, with exposure of the dentin, leading to pain, impaired function and aesthetics of the teeth, which can interfere with the individual's quality of life. Erosive dental wear has a multifactorial etiology, which makes prevention and treatment complex. Preventive strategies with an emphasis on the use of fluorides have limitations, therefore, the use of bioadhesive polymers in order to form surface protective films and increase the substantivity of the assets present in fluoride compounds has been investigated. Promising results were found in laboratory studies carried out by our research group using polymers, with emphasis on the polymethacrylate copolymer. Thus, in order to understand and characterize the preventive mechanism of action, in this study, we aimed to measure the effect of the association of solutions containing F (Sodium Fluoride NaF - 225 ppm F-) and/ or S (Stannous Chloride SnCl - 800 ppm Sn+2) and/ or AMC copolymer (2% Aminomethacrylate) in the anti-erosion potential of the acquired pellicle over enamel and dentin, in the face of erosive challenges, using a randomized cross-over in situ study. For this, twelve previously selected volunteers will install in the morning an intraoral device containing four specimens of bovine teeth (two enamel and two dentin) and wait 5 minutes. Afterwards, they will do the mouth rinse with 5 ml of one of the solutions to be tested (1 minute), and remain for 30 minutes without feeding using the palatal device, to allow the maturation of the acquired pellicle. Then, the specimens will be subjected to the erosive challenge with 0.03% hydrochloric acid, pH 2.3 for 3 minutes extra-orally. These steps will be repeated, implementing an erosive cycling, with five applications of treatment solutions, being: ultrapure water (negative control); F; S; AMC; FS; F associated with AMC; FS associated with AMC. Each solution will be tested in separate phases with the same volunteers, with a two days washout interval. After the end of the cycle, the specimens will be removed from the device and the substrate microbiopsy will be performed with 0.03% hydrochloric acid, pH 2.3 for 3 minutes. The measurement of the concentration of calcium and phosphorus will be performed by colorimetric method in a spectrophotometer in the collected acid samples. The data will be submitted to the appropriate statistical analysis, according to the normality tests, at a significance level of 5%. (AU)
