Abstract

In this project, the electrical impedance spectroscopy technique will be explored for the detection of Staphylococcus Aureus bacteria based on the variation of capacitance and resistance to the current flow among the interdigitated gold electrode tracks containing the specific antibody, in order to monitor the antibody-bacteria interaction. More specifically, the anti-S. aureus will be deposited among the gold tracks on the glass previously modified with the polyethyleneimine polymer (PEI), which will serve as an immobilization matrix, through the technique of self-assembled films layer by layer. The assembly of the PEI/anti-S aureus film will be characterized by impedance spectroscopy and atomic force microscopy (AFM). Detection will be performed with the inactivated S. aureus bacteria and selectivity tests will be conducted with strains of other inactivated bacteria. Data analysis will be performed, following models described in the literature, through changes in conductivity and dielectric permittivity that are reflected in the impedance and can be used to calculate the concentration of the species to be detected, as well as by principal component analysis component (PCA). At the end of the project, it is expected the formation of the student in the area and to obtain a prototype of a biosensor operating in the non-fararic mode for the detection of the bacteria in food samples.(AU)
