Systemic supplementation with Coenzyme Q10 in the periodontium of rats treated with nicotine: qualitative analysis of collagen fibers and quantitative analysis of fibroblasts

Abstract

The aim of this study will be to evaluate qualitatively the periodontal collagen fibers and quantitatively fibroblasts in the periodontium of rats treated with nicotine submitted to systemic supplementation with Coenzyme Q10. Sixty-four male rats (Wistar) will be divided into 4 groups (n=16). Throughout the experiment, animals will receive two subcutaneous injections of either 3mg/kg nicotine hemissulfate or physiological saline solution (PSS) with 12 h interval between them, iniciando 30 days before the start of systemic administration of coenzyme Q10 (CQ10) or vegetable glycerin. The solutions will be administered via daily gastric gavage, according to the groups: SS-S e NIC-S: daily gastric gavage with 1ml of vegetal glycerin; SS-Q10 e NIC-Q10: daily gastric gavage with 120 mg of CQ10. The euthanasia will be performed at 7 and 28 days after initiation of systemic administration with CQ10 or vegetable glycerin. The specimens will be collected and processed for histopathological analysis, qualitative analysis using the polarization method with Picrosirus-red and quantitative analysis of fibroblasts. After normality and homoscedasticity analysis, data will be submitted to the most appropriate tests with assuming 5% of significance (pd0.05).
