Formulation of experimental dentifrices containing nano particles of Beta-TCP functionalized with fluoride and stannous for the prevention of erosive tooth wear on enamel and dentin

Abstract

This study will formulate experimental dentifrices containing ²-TCP nanoparticles functionalized with fluoride and stannous and test them in the prevention of erosive tooth wear on enamel and dentin. For this, slabs of each substrate (4 mm × 4 mm × 2 mm), obtained from bovine incisors, will be acquired. The slabs will be flattened, polished, and randomly distributed into the 7 experimental groups (n = 10/substrate): 1. Negative control - toothbrushing with artificial saliva; 2. Stannous fluoride dentifrice (Gengiva Detox Deep Clean, Oral-B - 1100 ppm F-); 3. Placebo experimental dentifrice (without actives); 4. Experimental dentifrice with F- (1100 ppm F-); 5. Experimental dentifrice with F- and Sn2+ (1100 ppm F- and 3500 ppm Sn2+); 6. Experimental dentifrice with ²-TCP nano particles functionalized with F- (1100 ppm F- and ²-TCP); 7. Experimental dentifrice with ²-TCP nano particles functionalized with F- (1100 ppm F-, 3500 ppm Sn2+ and ²-TCP). Then, the specimens will be subjected to an erosion-abrasion cycle, consisting of 2 min immersion in 1% citric acid solution, followed by immersion in artificial saliva for 60 min, 6 times a day. The brushing will be carried out in a brushing machine, with soft toothbrushes and a suspension of toothpaste and artificial saliva, in the proportion of 1:3, 2 times a day. This procedure will be repeated for 5 days. After cycling, specimens will be analyzed as their surface loss (in ¼m) with an optical profilometer. The amount of KOH-soluble fluoride will also be evaluated (em mg/cm2). The pH, the potentially free fluoride, and the amounts of calcium, phosphate and tin in the toothpaste suspensions will be analyzed. Additional dentin specimens (n=5) will be subjected to 20,000 abrasion cycles to determine the potential abrasiveness of the toothpastes. Data will be statistically analyzed, considering a significance level of 5%. (AU)
