Influence of chronic consumption of western diet plus gamma-oryzanol on the morphology and function of renal and cardiac tissues in Wistar rats

Abstract

Important social, economic, and technological changes, resulting from the globalization process, have been impacting society's lifestyle. In general, this situation has influenced the population's eating habits, increasing the consumption of foods rich in sugars and fats, processed and ultra-processed, characterizing an eating pattern known as the western diet. The intake of this diet, accompanied by an increasingly sedentary lifestyle, physical inactivity, and genetic factors, characterizes obesity, a growing problem associated with several comorbidities, such as the development of kidney and cardiovascular diseases. The ³-oryzanol, the main component of rice bran, through works and studies, has demonstrated a positive action in the treatment of complications triggered by obesity. Aim: Thus, the present objective of this study is to investigate the preventive effect of the addition of ³-oryzanol in an obesogenic diet on the function and morphology of the heart and kidneys in Wistar rats. Material and methods: for the accomplishment of this study It will be used male Wistar rats (n = 40), 30 days old, from the Universidade Estadual Paulista (UNESP). The animals will initially be randomly distributed into four groups to receive a standard diet (SD) + water (n=10 animals), a standard diet with ³-oryzanol (DPG) (n=10 animals) western diet + 25% fructose in water, (DOF) (n=10 animals), western diet with ³-oryzanol + 25% fructose in water (DOFG) (n=10 animals) for 30 weeks. The characterization of obesity will be given by the assessment of nutritional profile according to the following parameters: caloric intake, body weight, and adiposity index. A 0.5% concentration of ³-oryzanol (Wako Chemical, Japan) will be added to the preventive diet and the treatment will be based on the work of Son et al. (2011). The cardiac function will be characterized by Doppler echocardiographic assessment. Renal function will be assessed by proteinuria and glomerular filtration rate. To evaluate the morphology, the organs will be stained for morphometric, collagen and inflammatory infiltrate analyses.(AU)
