Promising bioactive effect of a new nanoparticle loaded with hesperitin on bone metabolism: in vitro study

Abstract

Hesperidin is a flavonoid belonging to the flavanone subgroup and is found mainly in citrus fruits. In recent years this flavonoid has been widely studied due to its various properties such as antioxidant, anti-inflammatory, and beneficial effects on bone metabolism. On the other hand, interest in the use of nanoparticles such as nanoemulsions is growing more and more as a drug delivery system due to their structural design and outstanding biological properties. Thus, considering the beneficial effects of hesperetin associated with the characteristics of nanoemulsions, as well as the promising results obtained in our research group regarding the properties of hesperidin, the present study aims to evaluate the beneficial effect on osteogenesis of incorporated hesperidin in a new nanoemulsion as a locally controlled release system. Using Saos-2 human osteoblasts, non-cytotoxic concentrations of the nanoemulsion will be determined and its effects on proliferation as well as osteogenesis will be determined through the formation of mineralization nodules, alkaline phosphatase activity, organization, collagen maturation, and expression of regulatory genes of bone metabolism. Qualitative data will be analyzed descriptively and numerical data will be subjected to specific statistical analysis using the GraphPad Prism software.(AU)
