Abstract

This research project aims at the fabrication of microsensors for application in real-time studies of systems of biological interest. Two sensors for specific purposes will be developed. The first one will be employed to monitor the concentration of hydrogen peroxide released by keratinocyte-like cells, of HaCaT lineage, during processes of oxidative stress induced by 1,9-dimethyl methylene blue DMMB (10 nM), using radiation with a length of wave of 631 nm. The amperometric monitoring of hydrogen peroxide in cell culture medium will be performed with a gold microelectrode whose surface will be activated by the formation of nanoporous structures. The project will be carried out in collaboration with the team of Prof. Dr. Maurício da S. Baptista, from the Department of Biochemistry. The other microsensor will be applied in the monitoring of calcium ions release by bioactive dental restoration compounds. The sensor will have dual function: amperometric, to assure the sensor is always at the same distance to the surface pellets, and potentiometric, based on a selective membrane for calcium ions containing the ionophore ETH 1001. The detection of calcium ions released by the restoration compound over time will be performed with this device under different experimental conditions. The project will be carried out in collaboration with Prof. Roberto Ruggiero Braga. from the Department of Biomaterials and Oral Biology, Faculty of Dentistry, University of São Paulo. (AU)
