Effect of different post-cure times of printed resin composites on the mitochondrial metabolism and toxicity of human gingival fibroblasts

The aim of the present study is to evaluate the mitochondrial metabolism, the occurrence and type of cell death of human gingival fibroblasts exposed to restorative printed resins with different post-cure periods. Disc-shaped specimens of printed restorative resins (12 mm diameter and 2 mm thick for direct cytotoxicity and 12 mm diameter and 15 mm thick) will be prepared. To analize the indirect toxicity, the specimens will be immersed by 24h in pure culture medium, according to the ISO 10993-2016, to obtain the extracts (culture medium + products lixiviated from the resins) The cells will be exposed to the extracts for 3h, 6h and 24h. After this period, the mitochondrial metabolism of the cells will be evaluated by MTT test. The cell death will be evaluated after 24h of exposure by flow cytometry. For the direct toxicity, the 2mm thick discs will be positioned on 24 well plates and, the fibroblasts will be seemed on the discs. After 3h, 6h and 24h, the discs with the adhered cells will be transferred to a new 24well plates and the MTT test will be performed. The cell death by flow cytometry will be performed after 24h of contact with the discs. The obtained data will be plotted, and the statistical analysis will be performed, with a significance level of 5%.
