Abstract

Cystic adenoid carcinoma (CAC) is a rare tumor, accounting for about 1% of all head and neck malignancies and about 10% of all salivary gland tumors. Although they grow slowly, CACs have a progressive and infiltrative clinical course and can develop rapidly occurring metastases in high-grade disease. CACs are known to be difficult tumors to treat, especially because of their resistance to chemotherapy. With the advancement of Molecular Pathology, a plethora of potential therapeutic targets have been explored. Recent research has demonstrated epigenetic alterations in these tumors and provides important insights about histone deacetylase inhibitors (e.g. Entinostat) in these patients. In this current project we will perform Whole Exome Sequencing (WES) of four distinct groups of xenograft-derived ACCs: (1) untreated; (2) treated with Entinostat; (3) treated with cisplatin alone; and (4) treated with the combination of Entinostat and Cisplatin and (5) a group of ACCs from patients previously on therapy. Thus, we seek to broaden the genomic profile of this tumor beyond the already characteristic findings (tumors with or without rearrangement in MYB-NFIB) and also, evaluate the genetic changes as a result of the therapies used, trying to elucidate the role of epigenetic drugs in these tumors.
