Development of Calcium Silicate Endodontic Sealers: Evaluation of Physicochemical and Biological Properties

Abstract

As of the present moment, there are no endodontic sealers that meet all the ideal physicochemical and biological properties. The use of calcium silicate cements has the main advantages of bioactive properties and induction of hydroxyapatite formation; however, currently available commercial sealers exhibit high solubility. In this project, variations in the proportion of calcium silicate and radiopacifier will be tested in an experimental cement to enhance its physicochemical and biological properties. Tantalum oxide and zirconium oxide will be used as radiopacifiers. The employed proportions of calcium silicate will be 30%, 40%, and 50%. The liquid used will be propylene glycol containing 5% polyvinyl pyrrolidone. The studied properties will include setting time, pH, radiopacity, flow, volumetric change, and solubility. Radiopacity, flow, and solubility tests will be evaluated according to ISO 6876/2001 standards. Setting time will be assessed following ASTM C 266 - 2008 for hydraulic cements. For pH and volumetric change tests, acrylic teeth obturation will be performed with a single cone, and volumetric loss will be analyzed through computerized micro-tomography after 7 days of obturation. The water pH in which the teeth will be immersed will be measured with a pH meter at 03, 24, 72, and 168-hour intervals. Experimental cements showing promising results will undergo biological tests such as cell culture (cell viability test - MTT ASSAY) and subcutaneous implantation in rats. The obtained test data will be statistically compared (at a significance level of 5%) with the results of AH Plus Jet and Bio-C Sealer endodontic cements.
