Abstract Pleomorphic adenoma (PA) is the most common benign neoplasm of the salivary glands with a tendency to recurrence, recurrent transforming pleomorphic adenoma (RPA) and malignant transformation risks to carcinoma ex pleomorphic adenoma (CXAP). The presence of small non-coding RNAs, also known as microRNAs (miRNAs), can modulate the expression of genes related to the processes of tumorigenesis and carcinogenesis of the salivary glands. However, the transformation process of PA and RPA to CXAP is still uncertain and requires further studies. Thus, the objective of the present study is to evaluate the influence of miRNAs and potential targets in the malignant transformation of PA to CXAP, in the different stages of progression and subdivided according to histological types, in addition to investigating cases of AP recurrence. Fifteen PA, 15 RPA, and 15 CXAP paraffin embedded tissues will be evaluated. Total RNA will be obtained from all samples. Initially, the expression of 377 miRNAs will be assessed by quantitative real-time polymerase chain reaction (qRT-PCR), with TaqMan" Array Human MicroRNA A Cards v2.0 (Applied Biosystems") plates. After, the expression of the five major miRNAs will be validated by qRT-PCR, with TaqMan" (Applied Biosystems") probes. The prediction of potential targets will be performed by bioinformatics tools, in order to identify which genes - and their respective pathways - that are predictive of interaction with the miRNAs. The differences between groups will be assessed by Fisher's exact probability test or chi-square, t-test and Mann-Whitney. We believe that our results will contribute to expand the knowledge of the miRNA expression changes in the benign counterpart and whether or not they are conserved in their recurrence and in carcinoma, as well as possible new modifications of the recurrent and malignant tumor. | |
