Abstract

Parkinson´s disease (PD) is characterized by motor alterations resulting from nigrostriatal dopaminergic cells degeneration, which modifies the amount of dopamine in striatum unbalancing the motor circuit responsible for the normal movements. Genetic predisposition, aging and environmental toxins contribute to Parkinson's disease etiopathogenesis. In this context, toxins accumulation related to genetic variants of glutathione-S-transferases (GST) can injury dopaminergic neurons of substantia nigra, leading to PD. This study aims to analyze the frequency of genetic variant GSTP1 Alw 26I in PD patients; evaluate the association between GSTP1 Alw 26I polymorphisms and PD onset; and verify the association of GSTP1 Alw 26I variant and environmental toxins exposure in PD patients. One hundred and fifty patients with PD and 150 individuals without PD diagnosis (controls) will be studied. Allelic variants will be identified by polymerase chain reaction (PCR) and digested by Alw 26I enzyme for restriction fragment length polymorphism (RFLP) analysis, in 6% polyacrylamide gel. The statistical analysis will consist in parametric and nonparametric methods, according to the nature of the involved variables. The adopted significance level will be 0,05. (AU)
