Abstract

The ages-long relationship between humans and yeast has reached a new status over the course of the last decades. Swiftly, "Saccharomyces cerevisiae"-based products have shifted from traditional food and beverage goods to a plethora of new economic-relevant products applied in the most diverse sectors. The prosperity of the so-called yeast-based biotechnological platforms is intrinsically linked with the advancements in the genetic engineering field and the advent of metabolic engineering. In this context, the development of the CRISPR/Cas9 system for the efficient and rational genetic engineering of living organisms has completely revolutionized the way we obtain such biotechnological platforms. Together with other CRISPR-based approaches, such as the CRISPR/dCas9 system for genetic expression tuning, these techniques account for a potent genetic toolbox for the creation of suitable S. cerevisiae platforms for the most diverse industrial applications.
