Biochemical monitoring of SARS-CoV-2 VLP production stages using MIR spectroscopy and chemometrics

Abstract

Among vaccine platforms against SARS-CoV-2, the virus-like particles (VLP) hold advantageous immunological and safety attributes. Furthermore, there are barriers to the simultaneous and rapid monitoring of critical parameters throughout associated processes to ensure biodrug quality. This work is pioneering in advanced monitoring for producing SARS-CoV-2 VLP by the baculovirus/Sf9 insect cell system, using mid-infrared spectroscopy. It aimed to establish chemometric models based on spectra in offline attenuated total reflectance (ATR) and transmittance capture modes using partial least squares (PLS) and artificial neural network (ANN) for the biochemical monitoring (Viable cell density (XV), cell viability; glucose, glutamine, glutamate, lactate, NH+4 concentrations, and virus titer) of SARS-CoV-2 VLP production. Monocistronic recombinant baculovirus carrying genes of SARS-CoV-2 structural proteins and receptor binding domain spike protein (RBD) were used for VLP production by single or combined infections. Among assessed spectral segments, the complete spectral range was the most suitable. The results obtained by both models were mostly similar. However, ANN-created models stood out, providing the best performances, especially when the ATR capture mode was used. This becomes particularly evident in the XV, viral titer, and glucose parameters, which showed statistically significant differences, and lower relative error values of 27,5; 5,4, and 2,4 %, respectively.
