Gene and Protein Interaction among Components of the Inflammatory, Apoptosis and microRNAs Pathways in Gastric Disease and Gastric Cancer

Abstract

The persistent infection of the gastric mucosa by Helicobacter pylori (H. pylori), promotes a chronic inflammation, providing a microenvironment favorable for the appearance of atrophic gastritis with possible progression to gastric cancer. The stimulation to the immune and inflammatory response favors epithelial changes; which in turn stimulate apoptosis, the imbalance between the inflammatory process, death and cell proliferation being the main factor related to the progression of atrophic gastritis to gastric cancer. Deregulation in the gene and protein expression of molecules involved in these controlled events may contribute to the evolution of gastric diseases. The aim of this project: (I) evaluate the expression of miR-7, miR-let-7a, miR-25, miR106b, miR125a and miR155 microRNAs ; (II) To evaluate the expression levels (gene and protein) of apoptosis and cytokine regulatory molecules: Interleukin 6, IL-6 Receptor-±, TRAIL, DR4 AND DR5; (III) Correlate the expression of microRNAs, mRNA and proteins previously selected; (IV) To analyze the interaction miRNAs-mRNA-Protein with the presence of H. pylori and the bacterial virulence genes cagA, cagT and vacA. (V) Validate the results with functional assay in cell culture and bioinformatics. The development of this approach may open new perspectives for the characterization of factors involved in peptic diseases, gene regulation, as well as elucidation regarding the interaction between genes involved in gastric diseases, aiming at a better understanding of the pathophysiological mechanisms of these diseases. Together these data may suggest biomarkers and risk groups for progression to gastric cancer. (AU)
