Safety and efficacy for the prevention of dental erosion of a new cystatin derived from sugarcane (CaneCPI-5): in vitro, in situ and in vivo studies

Abstract

Recently, procedures involving "Acquired Pellicle Engineering" have been proposed by our group as a new strategy to control dental erosion. The incorporation of active substances in rinse solutions or gels for topical application could favor the ability of the acquired enamel pellicle to protect against erosion. Preliminary experiments of our group revealed that CaneCPI-5, a protein cloned from sugar cane and expressed in E. coli, was able to protect enamel against initial erosion in vitro. However, before products containing this protein can be broadly used to control dental erosion it is necessary to test their safety and additional experiments that more closely resemble the clinical situation must be conducted to attest its efficacy. Thus, this project will be developed in 3 steps, with the following general aims: 1) To evaluate the cytotoxicity of CaneCPI-5, in concentrations ranging between 0.025 and 1.0 mg/mL, in cultures of human fibroblasts; 2) To evaluate the protective potential of the modification of the acquired enamel pellicle with CaneCPI-5 (added to solution or gel) against erosion associated or not to abrasion in situ; 3) To evaluate alterations in the protein profile of the acquired enamel pellicle formed in vivo after treatment with solution or gel containing CaneCPI-5. With this project we expect to have enough evidence regarding the safety, efficacy for the prevention of dental erosion and mechanism of action of solutions and gels containing CaneCPI-5. This will allow that these products are marketed all over the world, constituting a new approach for the control of dental erosion. (AU)
