Abstract
Cardiorenal metabolic syndrome (CRMS) is a pathological condition associated with the development of cardiac and renal dysfunction due to obesity and associated complications, in which oxidative stress plays an important role in triggering the carbonylation of proteins The gamma-oryzanol (³Oz), the main phytosterol present in rice grains, has demonstrated potential antiadipogenic, hypocholesterolemic, anti-inflammatory and antioxidant action. Therefore, the objective of this study is to evaluate the preventive effect of gamma-oryzanol on the carbonylation of proteins CRSM-associated. Male Wistar rats (n = 48), with 21 days of age, will be used, coming from the biothermium of Universidade Estadual Paulista (UNESP). The animals will be distributed in four experimental groups (n = 12 animals / group). The G1 group will receive standard diet (SD), G2 diet standard + gamma-orizanol (SD + yOz), G3 diet rich in carbohydrates and fat (CFD) and G4 diet rich in carbohydrates and fat + gamma-oryzanol (CFD + yOz) for 30 weeks. Consumption of chow and water will be monitored daily and body weight weekly. At the end of the experiment and prior to euthanasia, the evaluations of systolic blood pressure, echocardiogram and 24-hour urine collection to assess proteinuria and glomerular filtration rate. Fat deposits will be collected to characterize obesity in plasma to measure biochemical, hormonal and inflammatory parameters and the kidneys and heart to determine antioxidant activity, carbonyl species and proteomics (carbonylated proteins). In addition, the concentration of yOz in renal cardiac tissues and plasma by high performance cormatography (HPLC) will be measured. Parametric data will be presented as mean ± standard deviation and comparisons between groups will be performed by analysis of parametric variance (ANOVA) and complemented by the Tukey post-hoc test. Non-parametric data will be presented as median and interquartile (p25 and p75) and comparisons between groups will be performed by Kruskal-Wallis and complemented by Dunn's post-hoc test.
