Abstract

Cutaneous aging is a natural biologic process, however it can be accelerated by several factor such as exposition to ultraviolet (UV) A radiation. Among the factors leading to aging, one can highlight oxidative stress and the enzyme catalase is one of the endogenous mechanisms to prevent this process by reactive oxygen species neutralization. In this sense, an additional supply of catalase could contribute to prevent cutaneous aging and related processes. Once aging is partially related to UV damage to DNA, topical application of DNA repairing enzymes, such as photolyase, is also interesting. Fotoliase is capable of reversing cyclobutane pyrimidine dimers (CPD) formed in the DNA with UV exposition. Nonetheless, catalase and photolyase as other biomolecules present low biological half-lives and skin penetration might be difficult owing to size and hydrophilicity. In this project we will investigate methods to improve catalase and photolyase stability and/or skin penetration aiming at promoting higher efficacy. Two approaches will be separately used, namely nanoencapsulation of the enzymes into Pluronic L-121 polymersomes and pegylation. These two systems will be investigated and compared to the free enzyme to evaluate catalase efficiency in reducing oxidative stress in vitro (pig skin) and ex-vivo (human skin) and to evaluate photolyase efficiency in repairing CPD DNA lesions.
