Effect of the extract of the leaf of Psidium cattleianum on the oral biofilm, enamel demineralization and on the protein expression of Streptococcus mutans

Abstract

Researching alternatives that could offer the maximum efficacy and the minimum side effects, natural products have been studied regarding chemical control of microbial biofilm. The aim of this study is to evaluate the effect of the leaf extract from Psidium cattleianum analyzing the oral biofilm formed after an in situ and its ability to inhibit the enamel demineralization and proteome analysis of S. mutans. For the proteome analysis, Streptococcus mutans C180-2 will be cultivated in brain heart infusion broth with PIPES buffer. After 24 h, the biofilm will be treated with deionized water (negative control), chlorhexidine 0.12% (positive control) or the plant extract during 1 minute. After the biofilm formation, the cells will be harvested and a lisate will be prepared. The proteins extracted will be dyed with dyes and submitted to two-dimensional electrophoresis with the Ettan® DIGE system Amersham Bioscience). The gels will be scanned trough the program Typhoon® 9400 and analyzed by the software De Cyder. In the in situ study, enamel blocks will be obtained, selected trough surface microhardness (SMH initial) and fixed in palatal devices of ten panels (four blocks for each panel). The experimental period will be of three phases of 14 days each, with an interval of seven days between them. The panels will drop a 20% sucrose solution and the treatment solution (water, chlorhexidine or the plant extract). After the experimental period, the oral biofilm will be analyzed regarding the number of total streptococcus, the number of S. mutans and the presence of carbohydrate alkali-soluble. In the enamel blocks, surface microhardness will be again analyzed (SMH final) to calculate the percent microhardness change (%SMHC) and the cross-sectional microhardness will be assessed to calculate the mineral loss (Z).
