Abstract

Parathyroid hormone (PTH) is an important calcium metabolism regulator and is known to induce bone anabolism in humans and rodents, when intermittently administered. The bone anabolism caused by PTH administration, can treat osteoporosis and to accelerate bone healing. Although most studies investigating PTH functions involve bone cells, other cells such as odontoblast, that also produce mineralized tissue (dentine), have been reported as having ability to express PTH receptor. This study is aimed at investigating the effect of intermittent hPTH(1-34) administration on gene expression (Col1a1: collagen, type I, alpha 1; Dmp1: dentin matrix protein 1; Dspp: dentin sialophosphoprotein; Alpl: alkaline phosphatase; Pth1r: parathyroid hormone 1 receptor, apoptosis and proliferation rates and alkaline phosphatase (ALP) activity of osteoblastic linage cells (MDPC-23). The effect of intermittent PTH administration on dentine formation will also be evaluated. Incisors, continuously growing teeth, will be used to investigate the effect of PTH on dentine apposition rate in mice, by using fluorescent markings. The results will be evaluated by statistical tests, and values at 5% level of significance will be considered. (AU)
