Dispersive Raman spectroscopy using optical fibers "Raman probe" applied to the diagnosis of neoplasia in the skin and prostate

Abstract

This research project aims to apply the dispersive Raman spectroscopy technique to identify the biochemical changes associated with neoplastic diseases in tissues of human skin in vitro and in vivo and prostate in vitro. The main goal is to obtain diagnosis information that allows one to identify changes in the tissue and to classify the altered tissues from the normal, using diagnostic algorithms based on multivariate statistics and the PCA and a model that considers the basic biochemical and morphological components of the tissues that occur in the observed spectrum, since biological tissues exhibit spectral signature of these biochemical components with high sensitivity and specificity. Using a dispersive Raman system with near infrared excitation (830 nm) coupled to a fiber optic cable ‘Raman probe’ or a microscope (Micro-Raman), one can obtain spectra with low sample fluorescence and minimized background generated by the Raman fiber, opening the possibility for in vivo studies in biological tissues and to correlate them with in vitro results, providing important information for the development of equipments to aid diagnosis of neoplastic lesions. The system consists of a spectrograph with semiconductor laser excitation (830nm), a ‘Raman probe’ that excites and collects the Raman signal of tissue, and the microscope, used to obtain spectra of the morphological structures of skin and correlated with biochemical changes. The project proposes the study of samples of skin and prostate cancer in vitro and skin lesions in vivo, analyzed through a diagnostic algorithm based on PCA and discriminant analysis and calculation of relative concentrations of major biochemical components of tissues, allowing the Raman technique to be used in the diagnosis of diseases.
