Abstract

The obesity pathophysiology involves a complex interplay between peripheral flags and the brain metabolism. This study has the hypothesis that the stimuli elicited by ingested nutrients results in cerebral perfusion alterations related to adipose tissue excess. Objective: Measure brain perfusion changes with SPECT (Single-Photon Emission Tomography) after stimulation with 75 grams of glucose and avaluate them by Statistical Parametric Mapping ® (SPM) and SISCOM (ANALYZE 10.0 ® ) softwares. Also will be determinated the serum levels of glucose, insulin, leptin, the basal metabolic rate by indirect calorimetry (IC) and body composition by bioelectrical impedance analysis (BIA). Finally, the cerebral perfusion changes and other data will be correlated to the obesity pathophysiology. Methods: A blind, randomized, controlled, and two groups of people, being the experiment subject control of yourself. One "O" group with ten obese (18 to 40 years, BMI between 30 and 40 kg/m2) and a "NO" group with ten non-obese volunteers (same age group, BMI between 18.5 and 24.9 kg/m2, living in similar conditions to the obese), with no morbidities. The subjects of both groups will be submited to nutritional status assessment, IC, BIA and Brain Magnetic Ressonance, being this last one for the analysis of the SPECTs by SISCOM. Subsequently all the patients will be randomized into subgroups. "O-1" and "NO-1" (cases) receiving an oral solution with 75 grams of glucose after 12 hours of fasting and "O-2" and "NO-2" (controls) receiving the same volume of water under the same conditions, in a crossover study. All assessed with SPECT and serum glucose, insulin and leptin before and after stimulation. (AU)
