Abstract

The hypothalamus is a brain region that gathers information on the body's nutritional status and governs the release of multiple metabolic signaling molecules such as insulin and leptin to maintain homeostasis. Obesity is associated with insulin and leptin resistance in the hypothalamus. However, despite it was recently demonstrated that the insulin resistance is associated with the increase in inducible nitric oxide synthase (iNOS) is, at least in part, mediated by S-nitrosation of proteins involved in insulin signal transduction: insulin receptor b-subunit (IRb), insulin receptor substrate 1(IRS-1), and Akt. The role of S-nitrosation of insulin and leptin signaling in hypothalamus is not clear. Thus the major aim of this study is to characterize the S-nitrosation of the proteins IR, IRS-1, Akt e da STAT3 in hypothalamus of obese rodents and investigate the relationship of this phenomenon with leptin and insulin resistance as well as with the control of food intake.
