CDNA and lincRNA microarrays: identification of expression patterns related to metastatization in oral squamous cell carcinoma

Abstract

Oral squamous cell carcinoma (OSCC) is the major subtype of malignant neoplasm of oral cavity, with more than 95% of tumors arising in this region. OSCC has been associated with a worst prognosis and high morbidity. At the moment, there is any available study adressing gene expression patterns in metastatic cells from OSCC, neither lincRNA expression profiles in OSCC. Comparison between cDNA and lincRNA expression profiles in primary tumors and metastatic cells in lymph nodes could provide new evidences regarding to metastatic progress in OSCC. The aims of this study is to find different expression patterns between primary tumor cells, and metastatic cells, in order to identify genes and lincRNAs that may be related to metastatization process in OSCC. In this study, ten cases of primary oral squamous cell carcinoma, and their matched compromised lymph nodes (LN) will be retrieved from patients that underwent surgical excision at AC Camargo Cancer Hospital. Through laser capture microdissection metastatic OSCC cells in LN will be separated for microarray analysis. cDNA and lincRNA microarrays will be performed and compared to obtain differentially expressed genes, and compared to tumors without LN commitment. The identification of differentially expressed genes will be done by applying the statistical test for analysis of microarrays (SAM Significance Analysis of Microarrays). To confirm changes in gene expression,it will be used RT-PCR for 10 differentially expressed genes. Protein expression analysis will be performed using immunohistochemistry on tissue microarrays.
