Evaluation of prostaglandin F2alfa effect on uterine involution of postpartum bovine and buffalo cows

Abstract

This study aim to evaluate the effect of a PGF2± synthetic analogue (chloprostenol sodium) on uterine involution of postpartum bovine and buffalo cows, as much as to verify its relation with 13, 14-Dyhidro-15-Keto- PGF2± (PGFM), estrogen and progesterone plasma concentration and gene expression of uterine PGF2± (FP receptors), estrogen and oxytocin receptor. For both, 30 pregnant pluriparous Houstein-Zebu cows and 30 pregnant pluriparous Murrah buffaloes will be used, with corporal condition score over than 3.0 (1 to 5 scale). Just will be included animals that present normal parturition without retained placenta. After parturition, cows and buffaloes will be divided into three treatment groups: CON (control; N = 10 of each specie) will receive 2 mL of saline i.m. on days 2 and 5 postpartum (day 0 = parturition); CLO2 (N = 10 of each specie) will receive 2 mL of chloprostenol i.m. on days 2 and 5 postpartum; and CLO15 (N = 10 of each specie) will receive 2 mL of chloprostenol i.m. on days 15 and 20 postpartum. Gynecological exams will be done on days 2, 7, 14, 21 and 28, with evaluation of uterine involution (scale 1 to 3) via transrectal palpation, uterine fluid accumulation (scale 0 to 3) by B-mode ultrasonography and the restart of ovarian activity postpartum (presence of dominant folicle and corpus luteum) by ultrasonography exam. Blood samples, for plasma PGFM, estrogen and progesterone determination, and samples of endometrial tissue, to quantify FP, estrogen and oxytocin receptor gene expression by qRT-PCR, will be taken every gynecological exam.
