Abstract

This work deals with the synthesis, characterization, cytotoxicity evaluation and DNA binding studies involving chelated compounds of the type [Pd(N-C)(X)(L)] and [PdX2(N-S)] {N-C = C2,N-bzox, C2,N-mbz, C2,N-bzan (bzox=benzaldehydeoxime, mbz = N-benzylidenemethylamine, bzan = N-Benzylideneaniline); N-S = 1-thiocarbamoylpyrazoles; L = thiourea-type ligands, X=Cl, Br, I, SCN}. The compounds will be characterized by elemental analysis, IR and NMR spectroscopies, ESI/MS mass spectrometry, thermogravimetry (TG) and single crystal X-ray diffraction methods. The citotoxicity of compounds towards normal cells and tumor cell sensitive and resistant to cisplatin will be determined by M.T.T. assay. DNA binding studies will be carried out involving the compounds with promising Resistance Factor (RF) and Therapeutic Index (TI) values aimimg at gaining information about the intensity and kinectics of such interaction. The influence of the compounds on the tertiary structure of DNA will be determined by their ability to modify the electrophoretic mobility of the plasmid DNA. Viscosity experiments and thermal denaturation of DNA will be also carried out depending on the acquisition of the described instruments in this project. (AU)
