Molecular characterization of Salmonella Typhimurium strains isolated from humans, food and animals in the Southeast and South regions of Brazil

Abstract

The infections caused by Salmonella spp. due to their high occurrence have a great impact on the economy and global public health. Specifically, S. Typhimurium has been reported as the most frequently isolated serovar in the United States, Canada, Australia and New Zealand, and the second most frequently isolated serovar in African, Asian, European and Latin American countries, including Brazil, between 2001 and 2007. Due to the great importance of Salmonella, many phenotypic and genotypic studies have been performed to better elucidate its epidemiology. However, there are just few studies that used molecular methods to type S. Typhimurium strains isolated in Brazil. In this project it will be studied 89 S. Typhimurium, being 49 isolates from humans (43) and food (6), between 1983 and 2010, from various cities of the Sao Paulo State and 40 isolates from animals of the South Region of Brazil. The S. Typhimurium strains will be molecularly type by Pulsed-field gel electrophoresis (PFGE), Multiple-locus variable-number tandem-repeats analysis (MLVA), Multilocus sequence analysis (MLSA) based on the sequences of two virulence genes (sseL e fimH) and two CRISPRs loci (Clustered regularly interspaced short palindromic repeat) and Multilocus sequence typing (MLST) based on housekeeping genes. Moreover, the presence of some virulence markers of the Salmonella Pathogenicity Island (SPI) I and II, and virulence-associated plasmids will be searched by PCR. The set of results to be obtained will contribute for a better characterization of S. Typhimurium regarding its diversity, pathogenic potential and may outline in detail the epidemiology of this serovar of great clinical importance in Brazil and worldwide. (AU)
