Abstract

Amazon deforestation threatens the genetic resources of many native species of this region. Among the endangered species is the genus Hevea, exclusively derived from the Amazon rainforest, Hevea brasiliensis is an emblematic species in the Amazon region, due to its historic role in the economic development of the states that comprise the Amazon. The rubber tree is the basis of the natural rubber industry. However, due to a strong effect on the restriction effect of the genetic basis, more than 95% of the cultivated areas in the world has rubber tree clones originated from only 5-10 trees. The genetic diversity of the species is still present in native populations in Amazon, but is threatened by the rapid pace of deforestation. The main objective of this project is to characterize the genetic diversity of tree rubber collected from different regions of the Amazon by means of molecular markers "single base polymorphism" (SNP). These markers will be developed from the sequence comparison of a database of rubber tree ESTs (Expressed Sequence Tags) with the ones from the results of the transcriptome of rubber tree (the next-generation sequencing). The SNPs will be developed employing a mass spectrometer MALDI- TOF (Sequenom). The variability data obtained will be used in the construction of a molecular map and in the study of population structure and linkage disequilibrium of populations representing the germplasm of the Amazon, currently present in genebanks and therefore available to breeding. All results of genetic variability, the linkage disequilibrium and population structure generated as well as genomic resources (SNPs) developed in this project will be useful for genetic studies and breeding programs of H. brasiliensis, aiming to increase rubber production. The results will also be useful for efforts to conserve the species of Hevea in the Amazon, it will help to take measures in order to preserve the genetic diversity of rubber and related species.
