Abstract

Malaria causes about 1 million deaths annually, being one of the major problems of Public Health in developing countries. Plasmodium vivax is the dominant specie causing malaria outside Africa and it is responsible for 25- 40% of malaria cases in the world. P. vivax malaria has been considered a benign disease, however it could be highly debilitating and, in many cases, fatal. Recently, the infection with P. vivax has been associated in humans with the acute respiratory distress syndrome (ARDS) and characterized by a decreased in the capacity of gaseous changes, an increase of the leucocitary activity and in inflammatory mediators in the lungs. The molecular basis of this lung dysfunction is poorly understood due to the large number of putative factors involved in the ARDS and to the hindrances of human studies. Murine models have been crucial for the comprehension of pathogenesis of severe malaria, cerebral malaria, placental and anemic syndromes. However, there is no adequate and reliable animal model to study ARDS associated malaria. This project aims to establish and to analyze a murine model, which reproduces the observed pathological characteristics in human ARDS, as well as to identify molecular biomarkers with potential importance for a precocious diagnosis and accurate prognostic of ARDS associated to the malaria. The establishment of this model will contribute for the understanding of pulmonary malaria injury, through the elucidation of the molecular mechanisms of this disease. Moreover, it will allow to address the development of novel diagnostic and therapeutic strategies for ARDS. (AU)
