Abstract

Studies have suggested that Diabetes mellitus (DM) causes changes in the energetic metabolism and oxidative stress, which constitute important factors triggering cardiovascular disorders. The use of antioxidants has raised interest in clinics due to their therapeutic properties on cardiovascular diseases and thus may prevent morbimortality in diabetic patients. In this sense, the study of the effect of resveratrol (3,5,4'-trihydroxystilbene) based on serum biochemical parameters, energetic metabolism, oxidative stress and SIRT 1 expression in the cardiac tissue of rats with type 1 DM is proposed. The study will include 32 rats, with body weight of ± 250g. The animals will be allocated to experimental groups (n=8): G1 (normal, control); G2 (normal, treated with resveratrol); G3 (diabetic non-treated); G4 (diabetic, treated with resveratrol). Experimental DM will be induced by means of intraperitoneal administration of streptozotocin (STZ), single dose (60 mg/kg body weight). Resveratrol will be administered, through gavage, at the concentration of 1.0mg/kg/day during 30 days. At the end of the experimental period, calorimetric parameters will be assessed; then, the animals will be anesthetized and euthanized. The serum will be employed for glucose assay and lipid profile. Samples (200mg) of cardiac tissue will be used for assays of protein and glycogen concentration, oxidative stress analysis, and activity of energetic metabolism marker enzymes and activity of respiratory complexes, besides SIRT1 expression by Western Blotting. Statistical design was completely randomized with 4 treatments and 8 replicates, according to analysis of variance (ANOVA). The significance level will be of 5% probability and F statistics will be significant when p<0.05. Tukey's tests will be employed for comparison between means, and Mann-Whitney for analysis of the results obtained by the Western blotting technique. (AU)
