Abstract

Aspergillus niger is a fungus, belonging to the Nigri setion, which is of high biotechnological importance. However, it has been described that this species has the potential to produce two groups of mycotoxins harmful to human and animal health: fumonisin B2 and ochratoxin A. In Brazil, the knowledge of the incidence and genetic characterization of isolates of A. niger producers and non-producers of mycotoxins, particularly fumonisin, is very restricted. There is an overriding need for studies that reveal the incidence of toxigenic A. niger in Brazilian agribusiness products, in order to understand the damage they cause and outline preventive control measures. The objective of this project is to isolate and identify Aspergillus section Nigri in grape and onion produced in Brazil, and to evaluate the presence of ochratoxin A and fumonisin B2 in these foods. Isolates belonging to the species A. niger will be investigated for their ability to produce ochratoxin and fumonisin. Once the producer and non- producer strains of these toxins mentioned are identified, they will be used to study genetic variation in orthologous gene clusters in those previously described for Fusarium verticillioides (fumonisin biosynthesis) and A. ochraceus (ochratoxin biosynthesis). Genes that are present in producer strains and are absent in non- producer strains will be used for the development of detection systems specific for strains of A. niger producers of ochratoxin and /or fumonisin. The development of a molecular detection method directly (without cultivation) for monitoring A. niger mycotoxin producers will also be studied. (AU)
