In vitro biological effects of copaiba oil (Copaifera sp.), calcium hydroxyde and MTA isolated or in association on dental pulp stem cells

Abstract

Direct pulp capping materials are widely applied in the clinical practice in order to maintain the dental pulp vitality. They must be biocompatible and biostimulatory, it means, they must modulate the tissue response to injury. The calcium hydroxide (CH), mineral trioxide aggregate (MTA) and the copaiba oil (Copaifera sp; COP) isolate present most biological characteristics of the ideal pulp capping material. However, when associated to each other they could present more bioactive effect. Then, this study aimed at to analyze the proliferation, differentiation and migration of dental pulp stem cells from human deciduous teeth (SHEDs) in response to substances leached from these materials isolate and associated to COP. SHEDs from the Laboratório de Pesquisas Básicas do Departamento de Dentística da FOUSP, after new characterization will be placed in contact with medium conditioned by these materials. The experimental groups will be as follows: G1:Control: cells grown in ideal culture conditions; G2: COP- cells grown in medium conditioned by COP; G3: CH- cells grown in medium conditioned by CH; G4: CH+COP- cells grown in medium conditioned by CH+COP ; G5: MTA- cells grown in medium conditioned by MTA; G6: MTA+COP -cells grown in medium conditioned by MTA+COP. The cell proliferation will be assessed by analysis of mitochondrial activity (MTT reduction), the cell differentiation by analyzing the expression of dentin extracellular matrix proteins ((BGLAP, DSPP, DMP1 e HSP-27) using quantitative real time PCR (qRT -PCR), alkaline phosphatase activity and mineralized nodule production (alizarin red), and cell migration by scratch test. After analysis, the data will be compared by statistical tests to be determined after the analysis of the distribution of the results in each study (pd 0.05). (AU)
