Tissue and biomechanics analysis of bones of rats fed high-fat diet: experimental study of the relationship between fat mass and bone loss

Abstract

The aim of this project is to evaluate the relationship between fat mass and bone tissue and bone loss. This project will be divided into two steps, being: 1) preliminary to investigate the effects of high fat diet in Wistar rats infant (21 days) during growth until sexual maturation stage. This step will consist of 2 groups: PAD (fed with standard diet) and HIP (fed with high fat diet) and 2) dependent on the results of Phase I, may be: "Study of the effects of early -fat diet on bone tissue of ovariectomized rats, and if this diet will be able to accelerate the deleterious effects on bone, caused by ovariectomy" or "Study of the effects of the return to standard diet after ovariectomy in rats in early intake of high-fat diet". These methodologies will consist of experimental groups with bone loss, using the ovariectomy model and with different exposure times and types of diet (high-fat or standard). Thus, to answer the questions raised in this study, the analyzes will be used: insulin tolerance, biochemical analysis of markers of formation/resorption (RANKL, OPG, osteocalcin and TRAP), liver tissue analysis, gene expression in the bone test (RT - PCR) of genes related to adiposity and osteogenesis, macroscopic analysis, bone densitometry, mechanical testing of trabecular and cortical bone, and microstructural analysis of micro- CT and scanning electron microscopy.
