Abstract

Crohn's disease (CD) and ulcerative colitis (UC) are the most proeminent types of inflammatory bowel diseases (IBDs). The pathogenesis of IBD is a multifactorial process which leads to an exacerbated immune response with an imbalance of proinflammatory cytokines, adhesion molecules as well as reactive oxygen species (ROS), thereby a tissue injury is established. Antioxidants are able to arrest free radicals generated by cellular metabolism or exogenous factors, preventing injuries and loss of cell integrity. Several studies have shown biological effects related to the rich diet with phenolic compounds, such as antioxidants, anti-inflammatory, antimicrobial activities as well as anti-carcinogenic. The apple (Malus domestica) has a large number of active compounds, denominated phytochemicals, which are related to chronic disease prevention. Apple extract may protect the DNA. In addition, it can reduce proliferation and invasion of tumor cells. Thus, the aim of this study is to evaluate the anti-inflammatory and antioxidant effects of the phenolic compounds from apple juice in chronic experimental colitis, using the TNBS model. In order to understand the biological machinery involved in this process, we will evaluate the gene expression of TNF-±, NF-kappaB, ICAM-1, iNOS, COX-2 and antioxidant enzymes such as catalase, Cu-Zn superoxide dismutase and Mn superoxide dismutase.
