Molecular characterization of Campylobacter jejuni and Campylobacter coli strains of diverse origins

Abstract

The genus Campylobacter belongs to the family Campylobacteraceae and currently comprises 25 species. The two most common species of clinical significance are Campylobacter jejuni and Campylobacter coli. The isolation of Campylobacter as a cause of gastroenteritis in humans in European and North American countries is very frequent. However, the isolation and the study of Campylobacter are not very common in Brazil, making it difficult to assess the importance of these species to as a cause of disease in humans and animals, as well as, to determine the impact of their presence in food and the environment. Furthermore, there are few studies that used molecular methods for typing C. jejuni and C. coli strains isolated in the country. The objectives of this project are to evaluate the genetic diversity, the pathogenic potential and the antimicrobial susceptibility of C. jejuni and C. coli strains isolated from humans, animals, food and environment in Brazil. A total of 126 C. jejuni strains isolated from humans (53), animals (40), food (29) and the environment (4), and 60 C. coli strains isolates from humans (10), animals (21), food (10) and the environment (19) during 16 years will be studied. The strains will be molecularly typed by pulsed-field gel electrophoresis (PFGE), sequencing of short variable region (SVR) of the flaA gene, analysis of the CRISPR locus (clustered regularly interspaced short palindromic repeats) by high resolution melting analysis (HRMA) and multilocus sequence typing (MLST). Moreover, it will be checked the antimicrobial susceptibility by the technique of minimal inhibitory concentration and the presence of some virulence markers by PCR. Taken together, the results to be obtained may contribute for a better understanding of the pathogenic potential, genotypic diversity and epidemiology of C. jejuni and C. coli strains isolated in Brazil.
