Abstract

The epidemic caused by the human immunodeficiency virus (HIV-1) is the most important emerging infection in the last decades. Despite advances in understanding the pathogenesis of the virus and the immune response against infection, there is no effective vaccine against HIV yet. Several lines of evidence indicate that neutralizing antibodies, CD4 + and CD8 + T lymphocytes play a major role in immunity against HIV. Vaccines that induce cellular response against HIV-1 developed so far, are mostly aimed at inducing CD8+ T lymphocyte responses. However, the role of specific CD4+ T responses in immunity against HIV-1, as evidenced by studies with infected patients and experimental vaccines have confirmed the importance of the development of a vaccine inducing these cell subtype. In previous studies, our group showed that immunization with a DNA vaccine in mice encoding multiple CD4+ T cells epitopes from HIV-1 was able to induce broad, polyfunctional and long-term CD4+ and CD8+ T cells specific responses. Although the results obtained with this vaccine formulation are quite promising, DNA vaccines have shown limited immunogenicity in humans. For this reason, in the recent years different platforms for vaccine production have been developed in order to transpose this barrier.An alternative that has been explored with success is based on in vivo targeting of antigens to dendritic cells (DCs). A sub-population of CD8+ DEC205+ DCs also express the DEC205 endocytic receptor. Several studies have shown that construction of an anti-DEC205 antibody fused to the antigen of interest is capable to induce strong antigen-specific humoral and cellular immune response. The aim of this project is to generate and produce a chimeric anti-DEC205 antibody encoding CD4+ epitopes of HIV and evaluate its immunogenicity in BALB / c mice. (AU)
