Evaluation of the TiF4 gel protector effect on eroded human dentin: an in situ study

Abstract

This study aims to evaluate the effects of TiF4 gel on the eroded dentin. In stage I (in vitro), 80 previously eroded human dentin slabs will be randomly divided into 8 groups (n=10): G1 - negative control (no treatment); G2 - placebo gel (water + thickener); G3 - 2% NaF gel at pH 7.0; G4 - 2% NaF gel at pH 1.5; G5 - 5.42% NaF gel at pH 7.0; G6 - 5.42% NaF gel at pH 1.5; G7 - 4% TiF4 gel at pH 1.5 and G8 - 4% TiF4 gel at pH 7.0. After treatments, samples will be submitted to a 10-day erosive cycling by alternating immersion in citric acid solution (0.05 M, pH 2.3, 2 min, 6x/day) and artificial saliva. After 5 and 10 days of cycling, profilometric analysis will be performed (wear and surface roughness). In stage II (in situ), 256 slabs of human root dentin (3x3x1 mm) will be randomly divided into 4 groups (n=16): C - negative control; P - placebo; NaF - NaF gel and TiF4 - TiF4 gel. For fluoride groups, concentrations and pHs are going to be defined after stage I. Cycling will correspond to 5 days of erosive challenges in situ (0.5% citric acid, pH 2.3, 6x/day). Samples will be evaluated under profilometry (n=16), scanning electron microscopy (SEM), energy dispersive x-ray spectroscopy (n=3) and used to determine loosely and firmly bound fluoride concentrations (n=13). Additional samples (n=16) will be analyzed by cross-sectional nanohardness and erosive solutions used during cycling, analyzed for free fluoride ions concentration. In stage III (cell adhesion), 12 slabs of human root dentin (4x4x1 mm) will be randomly divided into 4 groups (n=3), which will be determined after stage II results. NaF and TiF4 groups that present the best results in terms of surface loss will be selected. The negative control and placebo groups will be maintained. One-thousand cells will be plated onto each treated slab and after 24 hours, samples will be prepared for SEM. On the electromicrographs will be performed the count of cells adhered on the surfaces. Quantitative results of all steps will undergo homogeneity and normality tests to determine the proper statistical test to be used (± = 5%). (AU)
