Systematic evaluation of differential protein expression in microorganisms and zebrafish using the neuropeptide kyotorphin and derivatives

Abstract

One of the major clinical problems currently is the increased resistance of microorganisms against antimicrobial agents. It is known that much of this resistance is due to the presence of a rich and complex ecosystem formed by the adhesion of one or more microbial types called biofilm. Biofilms are associated with approximately 80% of all medical infections in the world, and are highly and adaptively resistant to antibiotics. The bioactive peptides have emerged in recent years as an alternative strategy for the treatment of antibiotic resistant bacteria. In the last years, it has been demonstrated that some antimicrobial peptides (AMPs) can prevent and / or eradicate bacterial biofilms independently of antimicrobial activity against bacteria in planktonic form. The kyotorphin (KTP) is an endogenous analgesic neuropeptide isolated in 1979 from bovine brain. Recently, studies have shown a pronounced antimicrobial activity for the derivatives of KTP opening prospects for this project. Based on these facts, one of the aims of the present project is evaluate if derivatives KTP can prevent beyond the bacterial growth, the formation of bacterial and fungal biofilms. In addition, we will investigate patterns in the differential expression of bacterial proteins, exopolysaccharide matrix and zebrafish of samples treated and not treated with the derivatives of KTP correlating with their influence on bacterial growth, biofilm development and citotoxicity. This project will offer a platform for the development and training of human resources at different levels, as well as consolidating the use of zebrafish for proteomic studies of microorganisms.
