Abstract

The present study will be carried out by Rubian Xtract Serviços Ltda, the company is incubated in the INOVA/UNICAMP and associated to business incubator from ESALQTEC/USP. Our aim is to characterize the bioactive compounds in the oleaginous complex, extracted from annatto seed (Bixa Orellana) via supercritical fluid process, their effects on obesity and its co-morbidities. The extraction will be carried out by green technique using Co2 in supercritical phase. The obtained oleaginous complex from annatto seed contains the bioactive of tocotrienol-geranylgeraniol, in addition to fatty acids. The above mentioned compounds will be determined by chromatographic techniques. Antioxidant capacity will be investigated using ORAClipophilic assay and the lipid peroxidation will be assessed by peroxide value tests and TBARS test. Citotoxicity will be evaluated in vitro by XTT method. Further, the biological potential will be assessed in an in vivo experiment of diet-induced obesity. Body weight, insulin resistance (GTT and ITT tests), lipid profile (total cholesterol and its fractions), inflammatory markers (cytokines), oxidative status (antioxidant enzymes), hepatic toxicity (ALT, AST and alkaline phosphatase) will be determined. Commercial products of tocopherol and tocotrienol will be used as control. According to previous results, the oleaginous complex from annatto seed from supercritical extraction may have synergic effect of tocotrienol and geranylgeraniol. Moreover, that complex may show a strong antioxidant, anti-inflammatory and hypocholesterolemic outcome, which acts on control of obesity and co-morbidities. Therefore, the product may be utilized on nutraceutical and nutracosmetics industries. (AU)
