Abstract

The stroke is the second cause of death worldwide, each year more than 20 million individuals are affected by this disease, 5 million of these are the death. Brazil is the leading cause of death. Polls show that increasing the use of numerous animal models valid in research seeking treatments for stroke functional. Numerous species are used, rats, rabbits, dogs, pigs, sheep and species of primates. A large number of studies have been done with stem cells from different sources in different animal models of spinal cord injury, and even therapeutic trials in humans, showing promising results. The stem cells ideal for the performance of transplants should be immunologically inert, from sources of easy access, have rapid expansion in culture, ability to provide long-term survival and integration in the host site as well as being suitable for transfection and expression of exogenous genes. The objectives of this research is to establish a model of stroke in rabbits (Oryctolagus cuniculus) to the use of therapy with stem cells and measured by computerized tomography, behavioral assessment, clinical parameters and histological examination the lesion induced. Thus we design the experiment, the induction of stroke in rabbits, the cell culture-change of the dental pulp, transduction with retroviruses, the analysis of gene expression in lacz exogenous transduced stem cells prior to implantation in animals, the preparation of stem cells for transplantation in animals of transduced cells, computed tomography, the behavioral assessment, evaluation and gross histology. We emphasize that the absence of studies using markers for the tracking of stem cells after transplantation into brain vascular lesions, the possibility of combined use of cell therapy in animals with vascular brain injury from trauma and various diseases in veterinary clinics are facts that justify such a search, so we must emphasize the importance of research on cell therapy always, to direct and indirect insertions of biotechnology for human and veterinary medicine.
