Abstract

In the last years, our research group has focused on the search line of investigation of different antioxidants found in the diet and their interactions with the chemotherapeutic cisplatin and doxorubicin, drugs widely used to treat various types of cancers. The aim has been to identify protective effects of antioxidants on the adverse effects induced in healthy cells by treatment with chemotherapy, and is important for this protective effect does not influence the activity of the drug on tumoral cells. In literature are described several strategies for treatment with the association of natural or synthetic compounds with benefits for the cancer patient. Recently, our laboratory was estabilished in the line of Nutrigenomics research, that advocates the use of tools of the molecular biology to investigate in greater detail the gene-diet interaction. The goal is to expand on molecular level, knowledge about the mechanism of action to the antioxidants. Thus, taking into account evidence from epidemiological studies that have shown the beneficial effects of diets rich in antioxidants against the development of neurodegenerative diseases, we believe the potential for inhibition of neurotoxicity of antioxidants in the diet could be extended to adverse effects that arise during chemotherapy. The relationship between antitumor chemotherapy with cisplatin and the development of sensory neuropathy due to damage induced in primary neurons has been confirmed in oncologic patients. The hypothesis of this project is that coenzyme Q10 can reduce the toxicity of the drug cisplatin in PC12 cells in culture. The neuroprotective potential of coenzyme Q10 will be evaluated by the inhibition of lipid peroxidation and by means of the parameters of analysis falls in single and double breats of DNA by the comet test and analysis of the "Cytoma" in binucleated cells. In addition, to identifying possible modulation in expression of the TP53 gene with the methodology of real-time PCR. (AU)
