Effect of hyperlipidic diet on the expression of related proteins via intracellular insulin in hypothalamus of female mice at the stages of diestrus and proestrus.

Abstract

Despite the frequent association of hyperinsulinism and abnormalities in the reproductive cycle, the precise mechanism underlying the effects of insulin upon LH production and secretion is not completely clear. The association between insulin resistance, inflammation and hyperandrogenism maybe caused by the action of environmental factors like a intra-uterine insult, obesity, sedentary lifestyle and a inadequate diet. The present study wants to analyze the regulation insulin signaling pathways by focused DNA microarray and Western blot techniques, in hypothalamus of mice C57BL6 non-pregnant females in two phases of the estrous cycle (diestrous and estrous), fed with high fat diet. The estrous cycle, serum concentrations of gonadal steroids and pituitary gonadotrophin, tissue expression and phosphorylation states of proteins involved in intracellular signaling of insulin in addition to the expression of cytokines IL-1beta and TNF-alpha will be analyzed.
